Lois Collier began her acting journey in 1938 with a minor yet credited appearance in "A Desperate Adventure," featuring Ramon Novarro and Marian Marsh. Her career flourished between 1940 and 1949, where she became known for her roles as the leading lady in several B-movies, frequently sharing the screen with western icons like Bob Steele, Tom Tyler, and Dennis Moore. In 1950, she ventured into science fiction, starring in the serial "The Flying Disc Man from Mars."

Often referred to as the Fourth Mesquiteer, Collier featured prominently in seven of Republic Pictures' "The Three Mesquiteers" films, taking on significant female roles. She also portrayed Carol in the daytime drama "Dear John," which aired on CBS during the 1930s and 1940s. On December 6, 1948, she became part of "You," a show broadcast on KMGM in Los Angeles.

In 1949, she co-starred in "City Desk," a compelling drama that delved into the dynamics of a newspaper newsroom. From 1950 to 1957, Collier transitioned primarily to television, where she played Mary, the supportive girlfriend and sidekick of the protagonist in the series "Boston Blackie," appearing in 58 episodes from 1951 to 1954.
