María Nela Sinisterra was born in Buenaventura, Colombia, a vibrant city nestled along the Pacific coast. After completing her nursing education at the IFA (Instituto Femenino La Anunciación) in her hometown, she ventured to Bogotá, the nation's capital, where she gained recognition by participating in the 2006 reality show Señorita Bogotá.

In 2008, at just 19 years old, María took a bold step and moved to Argentina, where she featured in the reality series De frente al miedo, produced in Buenos Aires for the Caracol network. Her creative talents flourished in 2011 when her original concept for a sitcom materialized into the show ¡Todas a mí!, in which she not only starred but also co-wrote all the episodes.

Her acting career continued to flourish with her role in the Spanish-Argentine horror film Penumbra, also released in 2011. The following year, she made her debut in Spanish cinema with the comedy Solo para dos, directed by Roberto Santiago, alongside the actor Santi Millán. In 2014, she appeared in the Colombian adaptation of Corazón de León, sharing the screen with prominent Colombian actors Marlon Moreno and Manolo Cardona.

Sinisterra also featured in the critically acclaimed Colombian drama Tiempo muerto, which garnered several awards in Argentina. In 2015, she collaborated with Colombian-American actor John Leguizamo in the dramatic film Perros, directed by Harold Trompetero, and later appeared in the horror-thriller Pacífico, filmed in Bahía Solano. Her journey continued with her role in Buscando el muerto arriba, showcasing her versatility as an actress.
